A GLP-1 dosage chart is only as good as its sources, so every row here comes from the manufacturer's current US label on DailyMed, read on 2026-09-24, or, for the one unapproved compound, from its published trial. The chart sets the products side by side: how each starts, how it steps up, where it stops, how often it is given and how long it lasts. A second table converts the injected doses into syringe units for powder vials. Nothing here is a plan for anyone; the labels set these doses for their indications, and a prescriber chooses among them.

Sources: the manufacturer labels listed at the foot of this page; the retatrutide row is the phase 2 trial (Jastreboff et al., NEJM 2023, PMID 37366315). "SC" is subcutaneous. The Rybelsus and Ozempic-tablet starting doses are, in the label's words, "not effective for glycemic control": they exist to start treatment. Zepbound's label says its 2.5 mg dose is for treatment initiation and is not approved as a maintenance dosage. Foundayo's molecule, orforglipron, is not a peptide.

What the chart shows that one label does not

The half-life sets the schedule. Read down the last column and the schedules follow: exenatide lasts hours and is injected twice a day, liraglutide about half a day and is injected daily, the three weekly molecules last five to seven days. Semaglutide's label adds that it remains in the circulation for about five weeks after the last dose.

Milligrams mean nothing across products. The same molecule appears at 2.4 mg a week injected and 25 mg a day by mouth on one label: 175 mg a week against 2.4, about 73 times more semaglutide by mass for the same weight-reduction indication. The oral label goes further and states that Rybelsus and Ozempic tablets "are not substitutable on a mg-to-mg basis", although both are semaglutide taken the same way. Between molecules, 2.5 mg of tirzepatide and 0.25 mg of semaglutide are both starting doses. No row converts into another, and the semaglutide to tirzepatide page reports that no label offers a conversion.

Every weekly label escalates slowly. Four weeks per step is the floor on all of them, and the labels give the same reason: to reduce gastrointestinal adverse reactions. Liraglutide steps weekly because it reaches steady state in days.

The injected doses as syringe units

The approved pens and vials are solutions of stated strength and are not measured in units. Powder vials are, and the arithmetic is units = mg ÷ mg/mL × 100 on a U-100 syringe. The concentrations below are common reconstitutions (5 mg/mL is a 10 mg vial with 2 mL; 10 mg/mL a 10 mg vial with 1 mL or a 30 mg vial with 3 mL; 20 mg/mL a 20 mg vial with 1 mL).

Any figure over 100 units is more than one 1 mL barrel holds, which is the arithmetic reason tirzepatide and retatrutide are usually worked at 10 to 20 mg/mL and semaglutide at 2.5 to 5 mg/mL. What the chart omits

The labels carry indications, contraindications, a boxed warning on thyroid C-cell tumours on every label here except Byetta's, missed-dose rules and dose adjustments that a chart cannot hold; each row is a summary of section 2 of its label, not a substitute for it. Paediatric doses appear only where they change the maximum. Products no longer marketed in the US (the weekly exenatide pen among them) are not listed because no current label was found for them. Retatrutide's figures are trial arms, not doses anyone has approved. Which dose anyone uses is decided with a prescriber.

What are the GLP-1 doses on the labels?

Weekly injections: semaglutide 0.25 mg rising to 2 mg (Ozempic) or 2.4 mg, with a 7.2 mg maximum (Wegovy); tirzepatide 2.5 mg rising in 2.5 mg steps to 15 mg (Mounjaro, Zepbound); dulaglutide 0.75 mg rising to 4.5 mg (Trulicity). Daily injections: liraglutide 0.6 mg rising to 1.8 mg (Victoza) or 3 mg (Saxenda). Twice daily: exenatide 5 or 10 mcg (Byetta). Why are oral GLP-1 doses so much higher than injected ones?

Because little of an oral peptide dose reaches the blood. The Wegovy label's tablet maintenance dose is 25 mg a day against 2.4 mg a week injected, about 73 times more semaglutide by mass each week. Foundayo (orforglipron) is not a peptide, and its milligrams are a different molecule's again. A milligram figure only means something for the product it was set for.

Which GLP-1 has the longest half-life?

Semaglutide, at about 1 week on its labels, followed by tirzepatide and dulaglutide at about 5 days. Liraglutide's is 13 hours and exenatide's (Byetta) 2.4 hours, which is why those are injected daily and twice daily. Foundayo's is 29 to 49 hours after an oral dose.

How long between GLP-1 dose increases?

At least 4 weeks for every weekly injection and 30 days for the daily tablets, on their labels. Liraglutide is the exception: Victoza and Saxenda escalate in weekly steps, and Byetta after one month. The labels give the reason as reducing gastrointestinal adverse reactions.

How many units is a GLP-1 dose?

Only a powder vial has units to count, and the number depends on the concentration: units = mg ÷ mg/mL × 100. At 5 mg/mL, 0.25 mg of semaglutide is 5 units and 2.4 mg is 48; at 20 mg/mL, 2.5 mg of tirzepatide is 12.5 units and 15 mg is 75. The approved pens and vials are solutions of stated strength and are not read in units.

Is retatrutide on the chart?

As a trial row only: it has no label. Its phase 2 obesity trial gave 1, 4, 8 or 12 mg once weekly, some arms starting at 2 mg, for 48 weeks. Those are study arms, not approved doses.
